Murray, 28, has a 2-0 record against Kyrgios, including victory in January's Australian Open quarter-final.
The winner will play Belgian David Goffin or France's Jeremy Chardy in the last 16.
Top seed Novak Djokovic takes on another Australian talent, 19-year-old Thanasi Kokkinakis, while nine-time champion Rafael Nadal plays Russian Andrey Kuznetsov.
In the women's draw, two-time champion and top seed Serena Williams faces former number one Victoria Azarenka.
