Blackpool took a surprise lead through Andrew Lyons in their first match since Mick McCarthy's departure from the club.
But Pelly-Ruddock Mpanzu scored twice to help Luton fight back to earn a 3-1 win, which puts Rob Edwards' side five points behind second-placed Sheffield United.
The result keeps Blackpool seven points from safety, with just five matches remaining of their season.
