Ashley Barnes scored the only goal of the game, converting from the penalty spot with 12 minutes remaining following a handball by Hatters defender Gabriel Osho.
Luton centre-back Tom Lockyer was then sent off for a second bookable offence, and the Clarets closed out the match for their 11th win in 12 league outings.
Elijah Adebayo had two good chances for the hosts in the first half, forcing Burnley goalkeeper Aro Muric into a save after 12 minutes but then failing to convert a cross from Pelly-Ruddock Mpanzu.
